Abstract

The purified extract of arum manis mango leaves contained flavonoid compounds by TLC methods and antibacterial activity against Gram-positive and negative pathogens. This research aimed to identify and determine the phytochemical content (mangiferin and flavonoids content) and antibacterial activity against other Gram-positive pathogens. Purification of ethanol extract of arum manis mango leaves by liquid-liquid fraction method. Determination of total flavonoid content and identification of mangiferin used respectively by aluminum chloride and Thin Layer Chromatography method. The disk diffusion method was used to assay antibacterial activity against S. epidermidis, and B. subtilis bacteria with the concentration of the sample such from 0.1, 0.5, 1, 1.5, and 2 mg/disk; meanwhile, against S. mutans at 62.5, 125, 250, 500, 750 and 1000 mg/disk. The data of total flavonoid content was analyzed by an independent T-test, while antibacterial assay and mangiferin identification by descriptively. The applied fractionation procedure could enrich the phenolic constituents of the arum manis mango leaf extract. Total flavonoid content shows a significant difference between purified extract and ethanol extract at 12,72 and 11,56 mgEQ/gram extract, respectively, that increased 1.1 times than of ethanol extract. The purified extract was identified a mangiferin at Rf 0.68, that similar to the mangiferin standard at Rf 0.67, and obtained the diameter of inhibition zone against S. epidermidis and B. subtilis at 8,1–15,9 mm, but no activity against S. mutans.
